    For – Lack of angiogenesis, inflammatory rich zones, low survival of transplanted cells, nutritional deprivation, and low differentiation of stem cells upon transplantation to the ischemic site are significant drawbacks in cell therapies for myocardial tissue regeneration. To overcome these limitations, we modified the intracellular signaling mechanisms of human adult stem cells by activating the delta opioid receptors (DOR, GPCR’s family). For the first time, we elucidate the role of DOR signaling on human adult stem cells. We found that the activation of DOR signaling enhanced the survival of hMSCs via the DOR/PI3K/AKT pathway with the suppression of pro-apoptotic markers Bax, Bad, and the upregulation of anti-apoptotic marker Bcl2 in deprived of nutritional conditions. It also increased the differentiation of hMSCs into cardiomyocyte-like cells via the NOTCH1 signaling pathway. Further, DOR signaling enhanced angiogenesis by VEGF secretion and the anti-inflammatory property of the hMSCs invitro.
